Human Topoisomerase II Beta Relaxation Assay Kits

These kits contain everything required for relaxation reactions. Including Human Topo IIb enzyme, the supercoiled pBR322 DNA substrate, the ATP, Assay and Dilution buffers as well as 10X GSTEB - our new stop and loading buffer.

Human DNA Topoisomerase II beta (Topo IIb) is a type II topoisomerase that can decatenate and relax supercoiled DNA. It is prepared by overexpressing in baculovirus-infected insect cells (Spodoptera frugiperda) and purifying it by methods developed in-house. The enzyme is supplied in Dilution Buffer.

Human Topoisomerase II is less efficient at relaxation than decatenation and ultra-high quality enzyme is required to catalyse the relaxation reaction.

1 U of Human Topo II will relax 0.5 µg supercoiled pBR322 DNA in 1X Assay Buffer in a 30 µL reaction volume in 30 minutes at 37°C.
