Human Topoisomerase I
Human DNA Topoisomerase I (Topo I) is a type IB topoisomerase which can relax supercoiled DNA. It is prepared by overexpressing the enzyme in baculovirus-infected insect cells (Spodoptera frugiperda) and purifying it by methods adapted from Stewart et al., 19961. The enzyme is supplied in Dilution Buffer.
1 U of topo I will relax 0.5 µg supercoiled pBR322 DNA in 1X Assay buffer in a total reaction volume of 30 µl in 30 minutes at 37°C.
This enzyme is supplied with 10X concentrated Assay Buffer and Dilution buffer which are also available separately.
Store at -80°C.
