The Human Topoisomerase II Alpha Decatenation Assay Kit provides all the essential components required to perform reliable decatenation and relaxation reactions. Each kit includes purified enzyme, catenated DNA substrates, a decatenated marker (for kDNA kits), Assay and Dilution buffers, and our 10X GSTEB stop and loading buffer.

Our newly developed BisCat DNA decatenation substrate consists of two supercoiled, singly linked minicircles of 2.3 kb and 2.6 kb. Type II topoisomerases efficiently decatenate this substrate, producing two supercoiled minicircle products that can be easily visualised.

Human DNA Topoisomerase II alpha (Topo IIα) is a type II topoisomerase that both decatenates and relaxes supercoiled DNA. The enzyme is expressed in baculovirus-infected Spodoptera frugiperda insect cells and purified using in-house protocols optimised for performance and consistency. The enzyme is supplied in Dilution Buffer for stability and ease of use.
