Fh8-tagged TEV protease

TEV protease is a sequence specific cysteine protease that recognizes the amino acid sequence ENLYFQ|G/S and cleaves precisely between the glutamine (Q) and glycine/serine (G/S) residues. The optimal temperature for cleavage is 30 °C but it also works as low as 4 °C

It's mode of action is like that of other cysteine proteases proceeds via enzyme adenylation, substrate binding, nucleophilic attack by the cysteine thiol on the peptide bond, and subsequent bond cleavage.

Inspiralis' TEV protease has an N-terminal 6-His-Fh8 tag allowing for traditional affinity purification or using a heavy-metal free process via a hydrophobic interaction purification matrix.

Key Features:

  • Fh8-tagged for heavy metal free purification
  • High sequence specificity
  • Equivalent or better activity with a variety of tagged proteins
  • Cleavage is 30 °C or at 4 °C

Applications:

  • Cleavage of purification or solubility tags from recombinant proteins
  • Directed cleavage of fusion proteins or modular proteins in enzyme engineering
